چکیده

The substrate inhibition of palmityl coenzyme A:carnitine 0-palmityltransferase (EC 2.3.1.-) by palmityl-CoA has been studied. Beside being a substrate for the enzyme, palmityl-CoA was found to behave as a competitive inhibitor for the second substrate (carnitine). The Ki was found to be approximately 3 X 10-e M, while the K, for palmityl-CoA was found to be approximately 10M5 M. Thus, the affinity of palmityl-CoA as an inhibitor for the enzyme was at least as high as its affinity as substrate. These results explain the pronounced substrate inhibition by palmityl-CoA. The K, values for the other substrates were found to be: Lcarnitine, 2.5 X lOA M; L-palmitylcarnitine, 4 X low5 M; and CoA, 5 X 10w5M. The possible physiological importance of the reaction kinetics of carnitine pahnityltransferase is discussed.

The effects of detergents of palmityl coenzyme A:carnitine palmityltransferase.

The effect of detergents on the activity of palmityl coenzyme A: carnitine 0-palmityltransferase (EC 2.3.1.-) has been investigated. A series of detergents (n-palmitylcarnitine, palmitylcholine, caprinylcholine, free fatty acids, deoxycholate, digitonin, Tween 20, Tween 80, and Triton X-100) have been found to stimulate the incorporation of L-carnitine-CH@H into L-palmitylcarnitine in the prese...

Adaptation of muscle to exercise. Increase in levels of palmityl Coa synthetase, carnitine palmityltransferase, and palmityl Coa dehydrogenase, and in the capacity to oxidize fatty acids.

The capacity of gastrocnemius and quadriceps muscles to oxidize palmitate, oleate, linoleate, palmityl CoA, and palmityl carnitine doubled in rats subjected to a program of treadmill running. The rate of palmitate oxdation by whole homogenates of, or the mitochondrial fraction from, leg muscles was twice as great per gram wet weight of muscle in the trained as in the sedentary animals over a wi...

Studies on acetyl-coenzyme A synthetase of yeast: inhibition by long-chain acyl-coenzyme A esters.

Long-chain acyl-coenzyme A (CoA) compounds (palmityl, stearyl, and oleyl) were found to be potent inhibitors of acetyl-CoA synthetase (ACS) of Saccharomyces cerevisiae strain LK2G12 from aerobic, but not from nonaerobic, cells. The effectiveness of the inhibitors of the aerobic enzyme was in the following order: palmityl-CoA < stearyl-CoA < oleyl-CoA. Short-chain acyl-CoA compounds (propionyl, ...

The effect of palmityl coenzyme A on glucose 6-phosphate dehydrogenase and other enzymes.

Palmityl coenzyme A was found to inhibit and inactivate both purified yeast and crude rat liver glucose 6-phosphate dehydrogenase. The yeast enzyme was inhibited 50% at a concentration of 0.0031 mm. The inhibition was strictly competitive and reversible with respect to glucose 6-phosphate and of the mixed type with respect to TPN. Bovine serum albumin and hydroxylamine both prevented and revers...

The localization of acyl coenzyme A-carnitine acyltransferases in rat liver cells.

The distribution of acetyl coenzyme A-carnitine acetyltransferase (EC 2.3.1.7) and of palmityl-CoA-carnitine palmityltransferase (EC 2.3.1) in subcellular fractions of rat liver has been studied. The distribution of carnitine pahnityltransferase in the different fractions obtained with differential centrifugation and with centrifugation in a steep sucrose gradient correlated closely with the di...
